Pushing the limits of sedimentary ancient DNA detection (LADA)

Sedimentary ancient DNA is an invaluable archive for reconstructing past changes in biodiversity in relation to past climate changes. Demonstrating that the DNA recovered from sediments is in fact ancient has only recently become possible by using shotgun sequencing and damage mapping. In this project, we will establish the methods of shotgun sequencing and damage mapping in our lab and apply them to the Fram Strait sediment core CAGE19-3-KH14. This core has been dated to approximately 243 kyr and thus represents a good opportunity to push the limits of the oldest detected sedaDNA in Arctic marine sediments. We will analyse the eukaryotic aDNA content of selected samples distributed throughout the core, verify that the DNA is in fact ancient and track changes in climate and biodiversity between ca. 20 to >240 kyrs.
